چکیده

The effects of sodium cyanate, ferf-butyl isocyanate, and benzyl isothiocyanate on carcinogen-induced neoplasia were studied in experiments in which the test compound was fed starting 1 week following completion of carcinogen administra tions. Under these conditions, all three test compounds exerted an inhibitory effect on the occurrence of 7,12-dimethylbenz(a)anthracene-induced neoplasia of the breast of Sprague-Dawley rats. In a second experiment model, sodium cyanate inhibited the occurrence of 1,2-dimethylhydrazine-induced neoplasia of the large bowel of female CF-1 mice. Thus, a new group of compounds has been identified which has inhibitory capacities against neoplasia when given subsequent to carcinogen exposure.

Effects of benzyl isothiocyanate on rat and human cytochromes P450: identification of metabolites formed by P450 2B1.

Naturally occurring isothiocyanates, such as benzyl isothiocyanate (BITC), are potent and selective inhibitors of carcinogenesis induced by a variety of chemical carcinogens. These effects appear to be mediated through favorable modification of both phase I and II enzymes involved in carcinogen metabolism. The inactivation of rat and human cytochromes P450 (P450s) in microsomes and the reconsti...

Chemoprevention by isothiocyanates.

Naturally occurring and synthetic isothiocyanates are among the most effective chemopreventive agents known. A wide variety of isothiocyanates prevents cancer in the rat lung, mammary gland, esophagus, liver, small intestine, colon, and bladder. Mechanistic studies have shown that this chemopreventive activity is due to favorable modification of phase I and phase II carcinogen metabolism, resul...

Benzyl isothiocyanate: double trouble for breast cancer cells.

An inverse association between dietary intake of cruciferous vegetables and cancer risk has been established for different types of malignancies, including breast cancer. The anticarcinogenic effect of cruciferous vegetables has been attributed to chemicals with an isothiocyanate (ITC) functional moiety. Research over the past three decades has provided extensive preclinical evidence for the ef...
